Thebeersampleswereanalysedforbiogenicaminesaccordingto
themodifiedGeigermethod[18].Theelutionprogramconsisted
ofagradientsystemwiththefollowingsolvents:(A)0.005M.
ammoniumacetatebuffersolutionadjustedtopH4.3withglacial
aceticacid;(B)methanol.Afterloadingthesample,analysisstarted
withamethanol(B)gradientinbufferAfrom65%to80%for
20min.andthenwithisocraticelutionof100%methanolfor
10min.Finally,thecolumnwasequilibratedwithanisocraticelution
of65%methanolinbufferAfor10min.
Theflowratewas1mL/min.Theeluteddansylamineswere
detectedbymonitoringtheUVabsorbencyat340nm.
Results
Twenty-sevensamplesofpopularPolishbeersweretested
byHPLCmethod.Eightbiogenicamines:monoamines
(TRP,PHM,HIS,TYR),diamines(PUT,CAD),
polyamines(SPD,SPM)andpolyamine-derivedsecondary
metabolites(PRP,PIR)wereinvestigatedinthebeer
samples.Thegradientelutionofmethanolinammonium
acetatebufferallowedelutionofalltheaminesinless
than30min.Inthetestedrangeofbiogenicamineconcentration
from0.2to10.0mg/Lthecorrelationcoefficient
betweenpeakareaandconcentrationofeachstandard
aminewashigherthan0.99foreverystandard
curve.Theaveragerecoveriesoftheaminesrangedfrom
81.6%to104.1%withthelowestvalueforspermine
